Specificity
This antibody recognises human Tissue inhibitor of metalloproteinases 1, also known as TIMP-1, Metalloproteinase inhibitor 1, Erythroid-potentiating activity or Fibroblast collagenase inhibitor. TIMP-1 is a 207 amino acid, ~23 kDa secreted glycoprotein containing a 23 amino acid signaling peptide and a single NTR domain. TIMP-1 is a metalloproteinase inhibitor that functions by formation of complexes with target metalloproteinases, binding to their catalytic cofactor leading to irreversible inactivation of the target metalloproteinase. TIMP-1 also functions as a growth factor and activates signalling cascades via CD63 and CD29 (Uniprot: P01033). In humans TIMP-1 also functions to promote growth and differentiation of erythroid progenitors. recognises both native and denatured TIMP-1, as well as TIMP-1 in and out of enzyme/inhibitor complex. This antibody is not known to be neutralizing.TIMP-1 is expressed by a wide range of cells including microvascular endothelial cells, smooth muscle cells, mesangial cells and some fibroblasts.Rabbit anti Humn TIMP-1 antibody has been used successfully for the localization of TIMP-1 in a rabbit model of hypertrophic scarring by immunohistochemistry on formalin fixed, paraffin embedded sections.
