Western blot analysis of various lysates, using Anti-Smad2 (phospho Ser245 + Ser250 + Ser255) Antibody [ARC53851] (A308749) at 1:2,000 dilution. 293T cells were treated by PMA/TPA (200 nM) at 37°C for 30 minutes after serum-starvation overnight. The secondary antibody was Goat Anti-Rabbit IgG H&L Antibody (HRP) at 1:10,000 dilution. Lysates/proteins were present at 25µg per lane. The blocking buffer used was 3% non-fat dry milk in TBST. Detection was with a ECL Enhanced Kit (RM00021). Exposure time: 60s.
