The VEGF (vascular endothelial growth factor) family and its corresponding receptors, VEGFRs (VEGF receptors), play significant roles in cancer. The VEGF family consists of several growth factors, including VEGF-A, VEGF-B, VEGF-C, VEGF-D, and placental growth factor (PlGF). These growth factors are secreted proteins that regulate angiogenesis and lymphatic vessels (lymphangiogenesis). Among them, VEGF-A is the most extensively studied member and has been linked to cancer progression and metastasis. The VEGFR family consists of three receptor tyrosine kinases: VEGFR-1 (also known as Flt-1), VEGFR-2 (also known as KDR or Flk-1), and VEGFR-3 (also known as Flt-4). These receptors are predominantly expressed on the surface of endothelial cells, which line blood vessels and lymphatic vessels. VEGFs play important roles within the growing tumour. VEGF-A stimulates the growth of new blood vessels, a crucial process for tumour growth and metastasis. It enhances the formation of abnormal and leaky blood vessels within tumours, facilitating the delivery of oxygen and nutrients to cancer cells. VEGF-C and VEGF-D primarily promote the growth of lymphatic vessels, contributing to the spread of cancer cells to regional lymph nodes and distant sites. VEGF signalling not only affects the vasculature but also directly influences tumour cells. It can thus promote cancer cell survival, proliferation, migration, and invasion. The overexpression of VEGF and activation of VEGFRs are frequently associated with an increased likelihood of metastasis and elevated levels of VEGF and VEGFR expression in tumours have been correlated with poor prognosis in various cancers. Consequently, targeting VEGF or its receptors has emerged as a therapeutic strategy in cancer treatment. Anti-angiogenic drugs have been developed to inhibit VEGF-mediated angiogenesis and are used in combination with chemotherapy in some cancer types.
