Early endosomes are essential components of the endocytic pathway in eukaryotic cells, playing a vital role in the sorting, trafficking, and processing of internalised cargo molecules. Structurally, early endosomes are dynamic, membranous compartments characterised by their tubular and vesicular morphology. These compartments possess a complex network of interconnected tubules and vesicles which undergo continuous fusion and fission events. Endocytic pathways involve the invagination of the plasma membrane to form specialised vesicles responsible for sequestering different cargo. For example, clathrin-mediated endocytosis internalises cell surface receptors, transporters and adhesion proteins into clathrin coated-vesicles, while caveolae-mediated endocytosis engulfs small molecules and extracellular vesicles within their own Cav1-containing caveosomes. Another pathway, known as macropinocytosis, leads to the formation of macropinosomes, large vesicles targeting extracellular fluid and macromolecules. Additionally, phagocytosis allows cells to internalise large particles including bacteria and viruses. In this process membrane extensions called pseudopodia surround these particles forming a phagosome. All these endocytic pathway-specific compartments fuse with early endosomes and can be identified by their membrane-bound protein composition. As a result, though not considered early endosome markers per, proteins such as clathrin heavy chain or caveolin-1 can be used to track vesicle dynamics and early endosome maturation. Early Endosome Antigen 1 (EEA1) is the best-established early endosome marker. It localises to the limiting membrane of early endosomes by interacting with phosphatidylinositol-3-phosphate. EEA1 also interacts with Rab5a, a small GTPase to regulate SNARE-docking and fusion events. Rab5a itself is considered a specific marker for early endosomes and is widely used in research applications to label and study these compartments. Other endosomal markers can be used in combination with EEA1 or Rab5a to study cargo trafficking between intracellular compartments. For example, Rab7 primarily localises to late endosomes and lysosomes but will transiently associate with early endosomes during their maturation. Further, Rab4a is involved in retrograde transport of cargo from early endosomes to the plasma membrane. In a similar manner, this marker localises to recycling endosomes which intersect with the early endosomal compartment.
