Immunofluorescent analysis of a rat cerebellum section stained with Anti-MeCP2 Antibody [5H12] (A104323), at a dilution of 1:1,000, in red, and co-stained with Anti-GFAP Antibody (A85419), at a dilution of 1:5,000, in green. The blue is Hoechst staining of nuclear DNA. Following transcardial perfusion of rat with 4% paraformaldehyde, brain was post fixed for 1 hour, cut to 45µM, and free-floating sections were stained with the above antibodies. Anti-MeCP2 Antibody [5H12] (A104323) specifically labels nuclei of neurons, while Anti-GFAP Antibody (A85419) stains the network of astroglial cells and projections of Bergmann glia.

Western blot analysis of different tissue lysates using Anti-MeCP2 Antibody [5H12] (A104323), at a dilution of 1:2,000, in green. The lanes contain: [Lane 1] protein standard, [Lane 2] nuclear extract of rat brain, [Lane 3] nuclear extract of mouse brain, and [Lane 4] cow cerebellum. The strong band at about 74kDa corresponds to the MeCP2 protein.
Immunohistochemistry analysis of a 4% PFA fixed paraffin embedded rat brain cortex section with Anti-MeCP2 Antibody [5H12] (A104323) at a dilution of 1:2,000 detected with DAB (brown) using the Vector Labs ImmPRESS method and reagents with citra buffer retrieval. Counterstained with Hematoxylin (blue). Anti-MeCP2 Antibody [5H12] (A104323) labels the nuclei of neurons with varying degrees of intensity. Note: this antibody performs well in testing with 4% PFA fixed tissues but is not recommended for immunohistochemistry on NBF fixed paraffin sections.
