Specificity
This antibody recognises human Flap endonuclease 1 also known as Flap structure-specific endonuclease, FEN1, Maturation factor 1 or DNase IV. FEN1 is a 380 amino acid ~45 kDa member of the FEN1 sub family of XPG/RAD2 endonucleases which has an important role in lagging strand DNA synthesis and base excision repair. It is hypothesised that FEN1 may play a role in the inhibltion of triplet repeat expansion. Two isoforms of FEN1 are generated by alternative initiation, isoform 1 against which rabbit anti human FEN1 antibody is generated and a truncated isoform FENMIT, lacking the N-terminal region and having a mitochondrial distribution, FENMIT also lacks DNAase activity. Initial results suggest that rabbit anti human FEN1 antibody does not recognize the truncated form of human FEN1.Fen1 is expressed in the nucleus of a wide variety of organ strusctures notably in those of the hematopoietic system including spleen, lymph nodes and spleen. recognises a band of ~45 kDa in whole cell lysates of HeLa and 293T by western blotting and has successfully been used for immunoprecipitation of FEN1 from HeLa cell lysates.
