Prior to reconstitution, centrifuge the vial at 5,000g for 3-5 minutes at room temperature. Reconstitute with appropriate volume of distilled sterile water to bring product to 1mg/ml concentration. After addition of distilled sterile water, mix by gentle tapping. Note: It is not recommended to vortex or vigorously pipette the sample.

Affinity ranking of different Rabbit Anti-BCMA Monoclonal Antibody clones by titration of different concentrations onto K562-BCMA (A) or NCI-H929 cells (B) at 4°C. Bound Rabbit IgG was detected in flow cytometry analysis. The Y-axis represents the mean fluorescence intensity (MFI) while the X-axis represents the concentration of IgG used.
ELISA plates were pre-coated with Recombinant Human BCMA Protein (Fc Chimera 6xHis Tag) (A318407), followed by pre-blocking with Human Anti-C11D5.3 Antibody (Grey bar) or Rabbit control IgG (Black bar), and then different Rabbit Antibodies were added to check the competitive inhibition of Human Anti-C11D5.3 Antibody. The clone DM3 exhibits the strongest inhibition (Red bar). This data indicated that DM3 bind to the same epitope as bb2121.
Immunoprecipitation analysis. Cellular overexpression lysates of HEK293F cells transfected with FLAG tagged human BCMA full length gene were pre-incubated with 6 different Rabbit Antibody clones and negative control IgG. The immunocomplexes were further pulled down by Protein A beads, fractionated, and blotted with Mouse Anti-FLAG Monoclonal Antibody.

(A) Anti-BCMA Antibody [DM6] - BSA and Azide free (A318678) does not bind to Jurkat cells that do not express BCMA. (B) A clear peak shift of Anti-BCMA Antibody [DM6] - BSA and Azide free (A318678) was seen compared to the control when incubated with BCMA-expressing MM.1S cells, indicating strong binding of Anti-BCMA Antibody [DM6] - BSA and Azide free (A318678) to BCMA. Antibodies were incubated at 2 µg/ml. .
