HEK293 cells were transfected with a construct including the N-terminal 608 amino acids of S. pyogenes CAS9 fused to GFP and stained with Anti-CAS9 Antibody (red). Transfected cells express the green fusion protein and bind the antibody in red, producing a yellow signal. Nuclear DNA in transfected and non-transfected cells is revealed with the blue DNA stain DAPI.

The lanes contain samples of: [1] Protein standards, in red, [2] non-transfected cells, [3] cells transfected with a construct encoding the C-terminal (814-1372) amino acids of S. pyogenes CAS9, and [4] HEK293 cells transfected with a construct encoding the N-terminal (1-608) amino acids of S. pyogenes CAS9. The 60 kDa and 68 kDa bands correspond to S. pyogenes CAS9 C-terminal and N-terminal constructs respectively.
