The lanes contain: [Lane 1] protein standard, [Lane 2] 10 µg of BSA, [Lane 3] 5 µg of BSA, [Lane 4] 2.5 µg of BSA, [Lane 5] 25 µl of Native Porcine GFAP Protein (A270561), [Lane 6] 12.5 µl of Native Porcine GFAP Protein (A270561), [Lane 7] 6.25 µl of Native Porcine GFAP Protein (A270561). The two protein bands in lanes 5-7 are alternate transcripts of the pig GFAP gene.
SDS-PAGE of Native Porcine GFAP Protein (A270561) was purified by a modification of the method of Leung and Liem. Cytoskeletal material was prepared by homogenization, detergent extraction and centrifugation to produce a pellet containing intermediate filaments and other stable protein complexes. This material was dissolved in 6M urea and separated by ion exchange chromatography on hydroxyapatite and then on a phosphate gradient on DEAE-cellulose to produce pure GFAP. The lanes contain: [Lane S] protein standard, [Next three lanes] indicated amounts of BSA, and [Last three lanes] three loadings of the GFAP fraction.
