The epitope for this antibody is located in the peptide HVPGGGNKKIETHKLTFREN, amino acids 362-381 of human Tau (NP_005901.2). This sequence is expressed in all known human Tau isotypes and is totally conserved in all mammals and other higher vertebrates.

Immunofluorescent analysis of cortical neuron-glial culture from E20 rat stained with Anti-Tau Antibody, at a dilution of 1:2,000, in green, and Anti-GFAP Antibody (A85307 | 1:5,000, in red. The blue is DAPI staining of nuclear DNA. Anti-Tau Antibody stains perikarya, dendrites and axons of neurons, while the Anti-GFAP Antibody labels astrocytes.
E18 rat brain cultures were grown for 10 days, fixed and immunostained with Anti-Tau Antibody (red), Anti-MAP2 Antibody (A85363 | green) and for DNA (blue). Anti-Tau Antibody stains the neuronal perikarya, dendrites and axonal process strongly and does not stain non-neuronal cells in these cultures. The Anti-MAP2 Antibody also stains neurons only, but only the perikarya and dendrites. As a result perikarya and dendrites appears yellow, since they contain both tau and MAP2, while axons appear red as they contain only tau.
Western blot analysis of tissue lysates using Anti-Tau Antibody, at a dilution of 1:2,000, in green,: [Lane 1] protein standard (red), [Lane 2] rat brain, [Lane 3] rat spinal cord, [Lane 4] mouse brain, [Lane 5] mouse spinal cord. Tau protein is expressed as up to 9 different isoforms of different molecular weight, and so appears as multiple closely spaced bands in the region of the blot from 48 kDa to 67 kDa with some higher molecular weight “big tau” Isoforms.

Immunohistochemistry analysis of a NBF fixed paraffin embedded human hippocampal section from an Alzheimer’s disease case with Anti-Tau Antibody [5B10] (A85415) at a dilution of 1:1,000 detected with DAB (brown) using the Vector Labs ImmPRESS method and reagents with citra buffer retrieval. Counterstained with Hematoxylin (blue). The Anti-Tau Antibody [5B10] (A85415) strongly labels nuclei and cell bodies in diseased neurons as identified by their morphology. Note: this antibody performs well in testing with 4% PFA or standard NBF fixed human and rat tissues.
