This antibody recognizes phosphorylated NF-H KSP sequences but not non-phosphorylated KSP sequences. In some species there is cross-reactivity with the phosphorylated KSP sequences found in the related neurofilament subunit NF-M.

Human cerebellar cortex fixed in formalin, embedded in paraffin and stained with Anti-NF-H Antibody using the ABC (avidin biotin conjugate) method. The section was counterstained with heamatoxylin-eosin (blue). Anti-NF-H Antibody stains prominent basket cell axons surrounding the large Purkinje neurons. Granule cell layer at bottom of image, molecular layer at top.
Western blot analysis of tissue lysates using Anti-NF-H Antibody (1:10,000 | green): [1] protein standard (red), [2] rat brain, [3] rat spinal cord, [4] mouse brain, [5] mouse spinal cord, [6] pig spinal cord, [7] cow spinal cord. Strong band at about 200-220 kDa corresponds to the major phosphorylated from of the NF-H subunit. A minor band at about 160 kDa is the non-phosphorylated NF-H. Smaller proteolytic fragments of NF-H are also detected in spinal cord preparations with Anti-NF-H Antibody.
