General Notes
This antibody can be used to verify the size of fusion constructs by western blotting, and to amplify the endogenous fluorescence of mCherry in transfected cells.
Immunofluorescent analysis of HEK293 cells transfected with mCherry-HA construct (red) and stained using Anti-mCherry Antibody (A270547), at a dilution of 1:2,000, in green. Nuclear DNA is visualised in blue using Hoechst staining. Anti-mCherry Antibody (A270547) reveals mCherry protein is expressed only in transfected cells which appear golden in color. Untransfected cells expressing no mCherry are not recognized by the antibody, so as a result only their nuclei are visible.

The lanes contain: [Lane 1] protein standards (red), [Lane 2] HEK293 control cells, [Lane 3] HEK293 cells stably transduced with a lentiviral vector expressing mCherry-HA construct, and [Lane 4] HEK293 cells transfected with mCherry-HA construct. The major band at about 30 kDa corresponds to mCherry protein.

Anti-mCherry Antibody (A270547) recognizes tdTomato and mCherry proteins revealing major bands at about 60 kDa and 30 kDa, but does not recognize GFP. The red band at 37kDa corresponds to GAPDH protein here used as a loading control.
Immunohistochemistry analysis of a formalin fixed paraffin embedded section of mouse brain transduced with Anti-mCherry Antibody (A270547) at a dilution of 1:10,000 detected with DAB (brown) using the Vector Elite ABC-HRP detection and reagents with citra buffer retrieval. Counterstained with Hematoxylin (blue). Anti-mCherry Antibody (A270547) specifically detected the soma and axons of oxytocin positive neurons in the hypothalamus. The TdTomato protein is very similar in sequence to mCherry so that our antibody strongly recognizes both proteins.
