Immunofluorescent analysis of a section of adult rat hippocampus stained using Anti-NeuN Antibody (A270544), at a dilution of 1:2,000, in red, and co-stained with Anti-MAP2 Antibody [4H5] (A85297), at a dilution of 1:2,000, in green. Nuclear DNA was revealed in blue using the DAPI stain. Following transcardial perfusion of the mouse with 4% paraformaldehyde, the brain was post-fixed for 24 hours, cut to 45 µm, and free-floating sections were stained with the above antibodies. Anti-NeuN Antibody (A270544) stains the nuclei of neurons in the hippocampus, while Anti-MAP2 Antibody [4H5] (A85297) stains the dendritic processes of neurons.
Mixed neuron and glia cultures stained with Anti-MAP2 Antibody (green) and Anti-NF-H Antibody (A85336 | red) and DNA (blue). Anti-MAP2 Antibody reveals strong cytoplasmic staining for dendrites and perikarya, which does not overlap with the Anti-NF-H Antibody which primarily binds to axons.

A band at about 280 kDa corresponds to the MAP2A and MAP2B proteins.

Immunohistochemistry analysis of a 4% PFA fixed paraffin embedded rat hippocampus section with Anti-MAP2 Antibody [4H5] (A85297) at a dilution of 1:1,000 detected in DAB (brown) following the ImmPress method with Anti-MAP2 Antibody [4H5] (A85297) strongly labels neurons, neuronal projections, and synapses. Note: this antibody performs well in testing with 4% PFA fixed tissues but does not stain NBF fixed human material effectively. We recommend clone Anti-MAP2 Antibody [5H11] (A85296) for human tissue immunohistochemistry.
SDS-PAGE of Anti-MAP2 Antibody [4H5] (A85297). The lanes contain: [Lane S] protein standard, [Next 5 lanes] 2µg of purified recombinant forms of various human MAP2 proteins. Note: D is recombinant full length MAP2D (559 amino acids) and C is recombinant full length MAP2C (471 amino acids), which is missing one of the microtubule binding domains of MAP2D and a more N-terminal 57 amino acid N-terminal sequence. Lanes P1, P2 and P3 show each recombinant segments of the human MAP2 projection domain, found in MAP2A and B but absent from MAP2C and D. P1: amino acids 233-684, P2: 712-1136, and P3: 1137-1588. All of the constructs run more slowly on SDS-PAGE than expected from their molecular size, probably due to their high content of charged amino acids. These constructs were used to make a series of mouse monoclonal antibodies. The lane marked BSA contains 2µg of bovine serum albumin, another gel standard.
