General Notes
Please note, Ki67 proteins are very unstable and only expressed in large amounts in situations where many cells are dividing. As a result of the very short half life of Ki67 there are usually numerous fragments visible on western blots running below the major 395 kDa and 345 kDa bands.
Synonyms
Antigen identified by monoclonal antibody Ki-67, Antigen KI-67, Antigen Ki67, MKI67, Proliferation marker protein Ki-67
High magnification confocal immunofluorescence image of Anti-Ki67 Antibody stained HeLa cells grown in tissue culture (green). The nuclei of rapidly dividing cells have clumps of Ki67 associated with nucleoli, while nearby quiescent cells express little or no Ki67. Cells are counterstained with Anti-Lamin A + C Antibody (red) which reveals the nuclear lamina, and DAPI reveals nuclear DNA in blue.
Western blot analysis of equal amounts of cell lysates using Anti-Ki67 Antibody (1:2,000 | green): [1] protein standard (red), [2] rapidly dividing HeLa cell cultures, [3] rapidly dividing HEK293 cell cultures. Strong double bands above 250kDa correspond to the two major Ki67 isoforms of apparent molecular weight of 345kDa and 395kDa. Smaller proteolytic fragments of these isoforms are also invariably detected on the blot.
