Principle of Assay
This kit provides a simple method for detecting succinate dehydrogenase (SDH) activity in animal and plant tissues, cells, and bacteria. The detection principle is based on SDH catalyzed dehydrogenation of succinate to produce fumarate. Phenazine dimethyl ester sulphuric acid (PMS) couples the dehydrogenation to the reduction of 2,6-Dichlorophenolindophenol (2,6-DCPIP). 2,6-DCPIP has an absorption peak at 605 nm. The reduction of 2,6-DCPIP is followed by measuring the change in absorbance at 605 nm, from which the SDH activity in samples can be calculated.
