Principle of Assay
Aspartate Aminotransferase catalyzes the transamination of alpha-ketoglutaric acid and aspartic acid to form glutamic acid and oxaloacetic acid. Oxaloacetic acid non-enzymically decarboxylates to form pyruvic acid. Pyruvic acid reacts with 2,4-dinitrophenylhydrazine(DNPH) to form 2,4, dinitrophenylhydrazone showing reddish brown in alkaline solution. Aspartate Aminotransferase activity can be calculated by measuring the formation of the colored 2,4, dinitrophenylhydrazone product at 500-520 nm.
